ok so I run two effects chains one for vocal and one for guitar.

I have two d/boxes, one for each. I understand that the d/box sends a balanced signal to a mixer and then to the house pa.

If I am practicing at home/or using a small amp for life shows/coffee houses/ the amp has a effects loop signal in the back...and a seperate mic hookup in the front.

Do I need to to run the guitar and mic to a d/box If I am not running them to a board, but rather to the amp directly.

thanks

I would not think so. The reason for the DI is as you said, to run to the board with a balanced signal.
We use DI's on the guitars and keys, but I don't use my effects loop, because it injects the effects right in between the preamp and the power amp. The signal strength will have to be set differently if the effects loop is used rather than the inputs. But, what the heck, try it and see.
